Some sources claim that isoleucine helps in the prevention/reduction of muscle damage post-exercise, however, the evidence is mixed. A systematic review in 2017, shows that there is no direct evidence of the positive effects of BCAAs on muscle damage, except in very specific conditions, which involve taking 200mg of BCAAs per kilogram of body weight daily for a long period of time.

On a positive note — claims suggesting that isoleucine may reduce fatigue during exercise may be true. A study published in the Journal of Nutrition suggests that an intake of BCAAs resulted in lowered ratings of perceived exertion and mental fatigue during a standardized cycle ergometer exercise. Apparently, BCAAs decrease the transport of tryptophan to the brain and decrease 5-hydroxytryptamine (5-HT) synthesis which, in abundance, are both responsible for central fatigue.

L-Valine (2000mg)

Also one of three branched-chain amino acids, valine is purported to provide numerous benefits, including energy enhancement, increased endurance, and muscle tissue recovery & repair.

Additionally, there are also claims that it may lower elevated sugar levels and increase growth hormone production.

A clinical study published in 2017, found that acute oral supplementation of BCAA’s at a concentration of 0.087 g/kg body mass was sufficient to increase the rate of recovery in isometric strength, CMJ height (countermovement jump), and perceived soreness compared to the placebo group.

Another clinical study published in 2018 in the scientific journal Bioscience found that acute supplementation of valine leads to a fatigue reduction during swimming exercise in rats. Whether that’s true for human subjects as well is still up for more research.

L-Glutamine (5,000mg)

Glutamine is one of the 20 naturally occurring amino acids in dietary protein and is also considered a conditionally essential amino acid that is elevated to essential status in periods of disease and muscle wasting derived from physical trauma. It’s usually sold in an isolated form, but Transparent Labs has decided to include it in Its formula.

Studies suggest that intramuscular glutamine levels play an important role in protein synthesis, as it may inhibit protein breakdown, as well as increase protein synthesis. However, such studies were conducted on rats, which means that it may not translate well to humans.

Glutamine is also an effective intestinal and immune system health compound, especially because it’s an important energy source or fuel for intestinal and immune cells. However, when taken up to two years, side effects such as fatigue, and loss of coordination may occur. As such, they should be used cautiously before or during activities where performance may require motor coordination.

BCAA’s may also cause minor side effects such as nausea, vomiting, diarrhea, and stomach bloating. In rarer cases, BCAA’s may cause high blood pressure, headache, or skin whitening.

Regardless of the supplement, it’s recommended that you ask your doctor or physician for advice, especially if you’re taking medication.
